Sample Preparation and Protein Purification

Immunoprecipitation is a technique used to selectively purify an antigen of interest present in a complex mixture. This is accomplished through the use of a specific antibody and a solid phase matrix to bring the antigen-antibody complex out of solution. This technique is often used to study the presence or absence of an antigen, the quantity of an antigen at a given time point, relative molecular weights of proteins, protein-protein interactions or post-translational modifications, or the rate of synthesis or degradation of antigens.

Antigens that have been recovered by immunoprecipitation can be utilized in a number of downstream applications, including functional assays and detection. One method of visualization of the purified antigen is Western Blotting. This method includes electrophoresis of the protein on a SDS PAGE gel followed by transfer to membrane and Western detection. KPL offers reagents for protein purification, immunoprecipitation, and protein analysis.

The ProteoSOLTM Tissue Extraction System lets you solubilize proteins in FFPE tissue samples for relative quantitative protein expression analysis, thereby enabling immunodetection of disease biomarkers.
